Abstract

This review focuses on current advances in researches of gasdermin family VSports手机版. The distinctive expression patterns and biological roles of members in this family were discussed. Most of them exhibit pore-forming activity on cell membranes and are executors for programmed cell death with cytokines release, and play roles in cancers and inflammation-driven diseases. Therefore, they can be used as potential therapeutic targets to treat related diseases. .

Gasdermin Family and cell lysis. All the members of Gasdermin family, except for DFNB59, are cleaved into two fragments and form pores on cell membrane by the released N-terminal domain, ensued by cell lysis. A) The N-terminal of GSDMA3 can be delivered to mitochondrion through translocase of outer mitochondrial membrane 70 (Tom70) receptor then interacts with mitochondrial chaperone TNF receptor-associated protein 1 (TRAP1) to promote production of reactive oxygen species (ROS). By opening the mitochondrial permeability transition (MPT) pores, membrane integrity is finally damaged then local inflammation and cell death follow. B) The N-terminal of GSDME permeabilizes mitochondrial membrane that results in cytochrome c release, and ultimately caspase-3 activation augments and apoptosis. C) Full-length GSDMB binds to the caspase activation and recruitment domain (CARD) domain of caspase-4 to enhance its enzyme activity. D) Pores formed by N-terminal of GSDMD also mediate release of IL-1β which is independent of membrane lysis and known as non-pyroptotic function for GSDMD. E) DFNB59 might take part in large proteins transport
